Decades after the apocalypse, Australia has become a radioactive wasteland, with the "Green Place of Many Mothers" being one of the last havens of fresh water and agriculture. While Furiosa and Valkyrie, two young girls, are picking peaches, raiders discover their location. Furiosa tries to sabotage the raiders' motorcycles but is captured and taken as a prize for Dementus, the leader of the Biker Horde.
Furiosa's mother, Mary, pursues the raiders to their camp, killing all but one, who manages to bring Furiosa to Dementus. Before he can reveal the location of the Green Place, Furiosa mortally wounds him. Mary then sneaks into the camp and rescues Furiosa, but Dementus tracks them down. In a desperate bid to buy time for Furiosa's escape, Mary gives her daughter a peach pit to remember her by, but Furiosa refuses to leave. Dementus forces Furiosa to watch as her mother is crucified. Haunted by his own family's death, Dementus adopts Furiosa against her will, hoping she will eventually lead him to the Green Place.
Some time later, Dementus besieges the Citadel, another settlement with fresh water and agriculture. However, his forces are repelled by the War Boys, the fanatical army of Citadel warlord Immortan Joe. Changing tactics, Dementus captures Gastown, an oil refinery that supplies the Citadel with gasoline, using a Trojan Horse strategy. At peace negotiations, Immortan Joe recognizes Dementus's authority over Gastown and increases its supplies of food and water in exchange for the Horde's physician and Furiosa, who has tattooed a star chart to the Green Place on her left arm to find her way home. Joe then imprisons Furiosa with his "wives" inside a vault. When Joe's son, Rictus, shows an interest in her, Furiosa devises a plan to escape. One night, Rictus breaks Furiosa out of the vault to assault her, but she evades him by using a wig made from her own hair and disappears.
Disguised as a mute teenage boy, Furiosa works her way up the ranks of Joe's men over the next decade. She helps build the War Rig, a heavily armed supply tanker designed to withstand raider attacks in the Wasteland. She plans to escape by hiding on the Rig when Joe sends his top driver, Praetorian Jack, on a supply run. Disillusioned by Dementus's cruelty, his lieutenant, The Octoboss, goes rogue and launches an air assault on the Rig. His Mortifiers slaughter the Rig's crew and destroy Furiosa's hidden motorcycle, but she and Jack team up to defeat them. Furiosa tries to hijack the Rig to return home, but Jack thwarts her attempt. Recognizing her potential, Jack offers to train her to escape if she helps him rebuild his crew. Furiosa agrees and becomes Jack's second-in-command, eventually earning the title of Praetorian. The two bond and plan their escape together, seeing an opportunity when Joe decides to attack Gastown, now mismanaged by Dementus.
Joe orders Furiosa and Jack to collect weapons and ammunition from the Bullet Farm, an allied mining facility. However, Dementus, who has already seized the farm, ambushes them. Furiosa and Jack barely escape, but Jack is captured and tortured to death. Furiosa, severely injured, escapes her chains by severing her own arm, sacrificing her star map to survive. A lone observer watches as she struggles back to the Citadel, where she and Joe's aide, The People Eater, form a strategy to counter Dementus. Luring Dementus into a trap at the Citadel, the War Boys crush the Horde in a 40-day battle.
Having lost her path home, Furiosa shaves her head again, replaces her arm with a mechanical prosthetic, and hunts down the fleeing Dementus. After an extended chase, she subdues him in the desert. The film's narrator, The History Man, recounts multiple versions of Dementus's death, finally revealing the true account: Furiosa imprisons Dementus in the Citadel and uses his still-living body as fertilizer to grow a peach tree from her mother's pit. Immortan Joe promotes Furiosa to "Imperator" and gives her command of a new War Rig. She meets Joe's five breeder wives in the vault where she was once imprisoned and shows them a peach from the tree. The night before another supply run, the "Five Wives" hide in Furiosa's Rig, setting the stage for their own escape.
